Presentation: Metabolomic and genomic prediction of 12 common diseases in half a million individuals from three national biobanks
We have measured nuclear magnetic resonance metabolomics from half a million blood samples from three national biobanks, and used those data to build risk scores that predict future risk of 12 diseases that cause the most morbidity in high-income countries. We compare the performance of these scores to polygenic risk scores and show that for most of the diseases considered metabolomics data is more effective at identifying high risk individuals. More than a quarter of individuals is at high risk for one or more of the seven diseases with good prediction and available interventions, suggesting that these scores could be useful in population health checks.

Poster: Genetic risk and changes in metabolomic risk over repeated measures
Metabolomic risk scores can complement polygenic scores to provide improved prediction of future risk of common diseases. Furthermore, because the biomarkers underlying these scores change in response to lifestyle and treatment, they can also chart an individual's risk trajectory over time. We have analysed 20,000 individuals with metabolomic profiles from samples taken 5 years apart, and show that both time points contribute to risk prediction, and are associated with positive changes in behaviors like weight loss and quitting smoking.

By systematically linking health outcomes to genetic and other molecular measurements at a previously impossible scale, they enable researchers around the world to carry out well-powered experiments. This workshop will offer a hands-on demonstration of how to work at this scale with clinical phenotype data, genetic risk scores, and Nightingale's metabolomics data.
